Why is it needed? Oil immersion is required for 100x objective lenses because, at high magnification, light rays bend (refract) as they pass from the glass slide into the air and then into the lens. This refraction reduces image clarity and resolution. Immersion oil has nearly the same refractive index as glass, allowing light to pass directly from the slide into the objective lens with minimal scattering. The result is a brighter, sharper image with improved detail—essential for observing bacteria, blood cells, and fine cellular structures under 1000x total magnification.
